I agree with Robert Beach that I thought there was no need to use a style because when I had tried reading MS Word styles with Jaws a few years ago, the only styles that Jaws read by default were the headings. I was able to change my settings in Jaws to make it ready all the styles, but many of the blind students I meet are not very advanced screen reader users. There are people on the list much more experienced than me and I appreciate anyone's information if this isn't correct, but it seems very unlikely to me that many screen reader users would change those settings.

On a related note that doesn't exactly address your question, a librarian who has more technical accessibility expertise than me had advice that I liked regarding accessibility and citation style. Since some citation styles such as APA require you to list the url, but accessibility guidelines advise meaningful link text, she suggested making the title the link, as you planned to do, and also including the url. However that was for a bibliography, where there's an argument that having the url text preserved over time is important. For a syllabus, I think it would be better to leave out the url text so it isn't extra clutter for everyone, especially blind users, because if the url is broken the professor is going to need to figure out what to do about that anyway; you don't have to preserve the url text for the potential that the document was printed and someone needs the url for historical record. I'm bringing this up as a consideration if any sticklers for citation style think the syllabus should have to follow citation style, which it does not sound like you've run into. (I'm always interested in citation style and accessibility, if anyone has other thoughts or points of view about that.)

In their syllabi, many of our faculty members want to turn their textbook titles into hyperlinks - to websites such as Amazon, where the students can buy the book.

We use MS Word documents for our syllabi. What's the best way of making these book-title/hyperlinks accessible?

My best guess is to use MS Word's Book Title style, and then make the text of the book title into a hyperlink.
For example, Disability Studies: An Interdisciplinary Introduction<https://www.amazon.com/Disability-Studies-Interdisciplinary-Introduction-Second/dp/1446280683/ref=sr_1_1?ie=UTF8&qid=1544639607&sr=8-1&keywords=Disability+Studies%3A+An+Interdisciplinary+Introduction>
Would a screen reader identify that as both a book title and a hyperlink?
Is there a better way to do it?

I would suggest only hyperlinking to the a descriptive link of the title of
the book, as you like APA etc.
and for print purposes include the hyperlink text but do not make it
clickable.

As to MS Word turning URLs into hyperlinks:
I think by default, MS Word does automatically turn URLs into hyperlinks, although you can change that in your settings.
Also, once it has turned something into a hyperlink, you can right-click to remove the hyperlink.
